While the airlines are simply publicly held company’s, all pilots must get FAA medicals. The FAA being a federal agency they will go back indefinitely and if there’s something on there they will find it. Further the airport your based at will also do a federal check so you can get an airport badge.
Long short if you’ve got something in your past you’re hoping won’t come to light it will.

You can provided its only one, it’s but recent and your record is clean otherwise (you also need to do well in training). It will also require some jumping through hoops to get your First Class Medical. The FAA will want all the details and will need to do their own research.
I recommend you consult with an AME (Aviation Medical Examiner) before you do much else besides an intro flight.

Usually a DUI can be overcome, but you had better have a very clean record besides that. Of course time is your friend with a DUI, the longer ago it was, the better.
